If a mother or father in the proband is known to possess the pathogenic variant recognized inside the proband, the risk for the sibs of inheriting the pathogenic variant is 50%. Due to minimized penetrance, sibs who inherit the pathogenic variant might or might not produce aHUS (see Penetrance).
